Pick Up the Canker Side Quest
To initiate the "Canker" quest, you must converse with Gules at Semine. You can easily pick up this quest during the wedding of Lord Semine and Agnes by speaking to Gules while he’s enjoying a drink before the ceremony. Alternatively, you can start the quest earlier by visiting Semine and speaking to Gules during his patrol. Inquire about his past with a group of bandits and offer your assistance in eliminating them. Canker, a former member of this group, becomes your first target.
Find and Kill Canker
Upon accepting the quest, Canker's location will appear on your map, situated north of Nebakov Mill. Be cautious as the path to his location is prone to ambushes by bandits. You can opt for stealth to avoid them, engage in combat, or simply flee. These random bandits are typically not challenging, but it's wise to conserve your health for the upcoming fight.
Canker's camp is nestled in the mountains, and it's easy to overlook as you need to pass through a narrow gap to reach it, similar to accessing the Hermit's house during the "Wedding Crashers" quest. Prepare for combat before entering, as you'll face several bandits alongside Canker.
The bandits may flee depending on the order you engage them, but you must defeat most of them. Fortunately, Canker won't escape, ensuring you can fulfill the quest requirement by killing him.
Finish the Quest and Return to Gules
After defeating Canker and his companions, loot Canker's body to collect the Light Mace, which serves as proof of your success. Feel free to take any additional items you find. Then, return to Gules to complete the quest.
Upon completing "Canker" in Kingdom Come: Deliverance 2, you must wait a full in-game day before you can pick up the next quest related to Gules' former bandit associates. Once the day has passed, speak with Gules to start "Handsome Charlie."
